Abstract

Inoculum size and N source were optimized for rapid production of xanthan by Xanthomonas campestris using fed-batch fermentation. N sources used were KNO3, NaNO3, NH4NO3, NH4Cl, (NH4)2SO4, urea, peptone and corn steep liquor. The fermentation period for production of xanthan by X. campestris decreased from 72 to 48 h using 30% (v/v) inoculum instead of the usual 10% level. Among the N sources tested in the medium having a C/N ratio of 28, the highest concn. of xanthan (1.9% (w/v)) with a viscosity of 10 000 cP was obtained with the KNO3 medium. Intermittent harvests operated for 3 cycles of 48 h each, using 30% inoculum, resulted in xanthan broth of high viscosity (9000 cP) with a concn. of 1.9% and a yield of 68% (w/w). It is concluded that KNO3 is a well suited N source for the medium used for rapid production of xanthan.
