Additional Information: Soybean (Glycine max) belongs to Papilionaceae is an important commercial crop highly valued for it rich fibre, protein and isoflavones content. Efficient methods of in vitro regeneration and callus cultures was established. A maximum of 8 shoots per explant were obtained from cotyledonary node explants on MS medium containing 0.2 mg/l thidiazuron. Further growth and shoot buds elongation was achieved in presence of 0.5 mg/l GA3. The elongated shoots were successfully rooted on MS basal medium within a month. Similarly callus cultures were established from leaves on MS medium containing 2.0 mg/l BA and later on 2,4 D (2.0 mg/l) and kinetin (0.5 mg/l) containing medium. In vitro stress conditions were created by incorporating the PEG into the medium and callus was inoculated onto the same to optimize the conditions and biomass was calculated at regular intervals. The isoflavones profile in both ex vitro and in vitro cultures was analyzed by HPLC. All the three isoflavones viz., daidzein, genistein and formanitin were found in soaked seeds samples and callus cultures.
