Additional Information: There has been considerable interest in the development and the production of biodegradable polymer to solve the current problem of pollution caused by the continuous use of synthetic polymer of petroleum origin, because their persistence in the environment causes deleterious effects on wild life and on the aesthetic qualities of cities and forest. So alternative materials to petrol-based plastics were searched. Many polymers were proposed and tested for their possible industrial application and their biodegradability eg: cellulose, starch, blends of those with synthetic polymers,polylactate, polyesteramide and polyhydroxyalkanoates (PHA). PHA gained particular interest since they have shown to be biodegradable and ion compatible. PHA is accumulated as a inclusion bodies in Bacteria, where there is an essential growth limiting component such as nitrogen, phosphate, sulfur, oxygen in the presence of excess of carbon. PHB is the best-known member of PHA with an enormous application.With this background the following objectives i.e. isolation of PHA from soil,characterization, production, extraction, purification and structure analysis by FTIR,was studied. Also the Pha C genes, which codes for synthase enzymes were studied. The organism was observed to produce PHA and was identified as Gram positive rods. The morphological and taxonomical evidence shows the organism belongs to the genera Bacillus circulans based on Bergey’s manual of determinative Bacteriology (volume 2,Peter. H.Smeath, Nicholus. S.mair, John.G.holt)). It is found suitable to grow utilizing Galactose as a sole carbon source at pH 7.2 at 37o C at 200 rpm. The organism is also found to contain Pha C enzyme that are important in PHA biosynthetic pathway and is a potent producer of PHA.
